Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, built from 44 amino acids. Its sequence follows the natural human GHRH(1-44) backbone, with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification blocks recognition by dipeptidyl peptidase IV, the enzyme that rapidly truncates the native hormone in circulation. The result is a molecule with a substantially longer plasma residence time than unmodified GHRH, which makes it practical for clinical and laboratory study.
Receptor-level activity begins when the peptide binds the GHRH receptor, a class B G-protein-coupled receptor found on pituitary somatotroph cells. Occupancy triggers Gs-mediated activation of adenylyl cyclase and a rise in intracellular cyclic AMP, which in turn promotes synthesis and pulsatile release of growth hormone. Because the compound acts upstream of the growth hormone axis rather than supplying hormone directly, its effect depends on intact pituitary function. Binding studies in cell culture and animal models have established this pathway; the detailed kinetics of receptor recycling in humans remain less well characterized.

Identity and purity are judged through a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact peptide from truncated, oxidized, and deamidated variants, and the resulting peak-area percentages yield a purity figure. Electrospray ionization mass spectrometry confirms the expected molecular mass and can expose unanticipated modifications. Amino acid analysis and peptide mapping support sequence fidelity, while water content, pH, sterility, and bacterial endotoxin testing describe the physical and microbiological attributes of a finished lot.
Regulatory position depends on jurisdiction and on the form in which the material is sold. A branded product holds approval in the United States for a defined indication, and prescribing is confined to that label. Material marketed for laboratory research is not evaluated for human use and carries no such clearance. Independent verification therefore rests on certificates of analysis, third-party testing, and documented chain of custody. The substance also appears on the World Anti-Doping Agency prohibited list within the category covering growth hormone-releasing factors.
Lyophilized material is typically held under refrigeration between two and eight degrees Celsius, shielded from light and ambient moisture. Peptides of this size adsorb to glass and plastic, so working procedures often call for low-binding containers and as few transfers as possible. Absorbed water during weighing shifts the apparent mass of a sample, and controlling room humidity reduces that source of error. Once dissolved, solutions are kept cold and used within the interval printed on the accompanying label or certificate. Degradation accelerates markedly in dilute aqueous form.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, composed of 44 amino acids. It was designed to retain the biological activity of the native hormone while resisting rapid enzymatic degradation. The compound is classified as a growth hormone secretagogue and belongs to the broader family of hypothalamic releasing factors. In research and clinical settings, it is studied for its ability to stimulate pituitary growth hormone release. Its structure includes a modification at the N-terminus that contributes to an extended half-life relative to native growth hormone-releasing hormone.
Tesamorelin binds to growth hormone-releasing hormone receptors on the surface of pituitary somatotroph cells. This binding activates adenylate cyclase, raising intracellular cyclic AMP levels and triggering the release of growth hormone into circulation. The elevated growth hormone then stimulates hepatic production of insulin-like growth factor 1. Because the effect is mediated through the endogenous axis, secretion remains subject to feedback regulation. This distinguishes it from direct growth hormone administration, which bypasses pituitary control entirely.
Clinical investigation has focused on HIV-associated lipodystrophy, a condition in which antiretroviral therapy contributes to abnormal fat distribution. Excess visceral adipose tissue accumulates in the abdomen while peripheral fat may be lost. Tesamorelin was evaluated for reducing this visceral fat depot, with trials measuring changes in abdominal fat by imaging rather than by body weight alone. The rationale rests on the known lipolytic effects of growth hormone. Effects on visceral fat are documented, while long-term outcomes regarding cardiovascular risk remain less clearly established.
Tesamorelin acts on the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in intracellular cyclic AMP, which in turn opens ion channels and raises calcium concentrations, leading to release of stored growth hormone into the bloodstream. Because the peptide works through the same receptor as the body's own GHRH, the resulting secretion follows a pulsatile pattern rather than a continuous elevation. The N-terminal modification slows enzymatic breakdown, so the signal persists longer than it would with the unmodified hormone.
Growth hormone released from the pituitary stimulates the liver and other tissues to produce insulin-like growth factor 1, a stable circulating protein that serves as a practical marker of activity. Clinical studies therefore track IGF-1 concentrations alongside the hormone itself, and they commonly measure body composition with imaging rather than relying on body weight alone. Visceral adipose tissue, the fat surrounding abdominal organs, is quantified by computed tomography in the studies that supported approval. Adverse effects reported in trials include injection-site reactions, joint pain, and increases in blood glucose, which is why monitoring accompanies use.
Questions remain about how much of the observed fat reduction reflects direct GHRH-receptor signaling versus the downstream growth hormone and IGF-1 surge. It is also unclear whether the compound produces meaningful benefit in populations without lipodystrophy, since trials in cognitive impairment did not reach their stated goals. Long-term effects on glucose metabolism and on cardiovascular outcomes are not fully characterized. Published work generally describes effects on surrogate markers rather than on hard clinical endpoints, and independent replication of some findings is limited.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
The study of catalysis and catalytic reactions is very important to the field of physical organic chemistry. A catalyst participates in the chemical reaction but is not consumed in the process. A catalyst lowers the activation energy barrier (ΔG‡), increasing the rate of a reaction by either stabilizing the transition state structure or destabilizing a key reaction intermediate, and as only a small amount of catalyst is required it can provide economic access to otherwise expensive or difficult to synthesize organic molecules. Catalysts may also influence a reaction rate by changing the mechanism of the reaction.
Dry ice is the solid form of carbon dioxide. It is commonly used for temporary refrigeration as CO2 does not have a liquid state at normal atmospheric pressure and sublimes directly from the solid state to the gas state. It is used primarily as a cooling agent, but is also used in fog machines at theatres for dramatic effects. Its advantages include lower temperature than that of water ice and not leaving any residue (other than incidental frost from moisture in the atmosphere). It is useful for preserving frozen foods (such as ice cream) where mechanical cooling is unavailable. Dry ice sublimes at 194.7 K (−78.5 °C; −109.2 °F) at Earth atmospheric pressure. This extreme cold makes the solid dangerous to handle without protection from frostbite injury. While carbon dioxide is not toxic at normal atmospheric concentrations, dry ice can create dangerous CO₂ levels in poorly ventilated spaces. The outgassing from it can cause hypercapnia (abnormally elevated carbon dioxide levels in the blood) due to a buildup in confined locations.
=== Historical definition === Sublimation is historically used as a generic term to describe a two-step phase transition – a solid-to-gas transition (sublimation in a more precise definition) followed by a gas-to-solid transition (deposition). (See below)

The event that precipitated the Cold War in continental Asia was the resumption of the Chinese Civil War (1927–1949) fought between the anti-communist Kuomintang and the Chinese Communist Party. After military defeat exiled Generalissimo Chiang Kai-shek and his Kuomintang nationalist government to Formosa island (Taiwan), Mao Zedong established the People's Republic of China on 1 October 1949.
Kyle Hardingham (born 1 September 1988) is a former professional Australian rules footballer who played for the Essendon Football Club in the Australian Football League (AFL). Hardingham was selected by Essendon with the seventh pick in the 2010 AFL Pre-Season Draft. Originally from East Fremantle in the WAFL, he represented Western Australia in the state game against South Australia in 2009. He made his AFL debut against North Melbourne in round 17 of the 2010 AFL season, kicking four goals, helping Essendon achieve an upset victory by 3 points. The following Friday night, he kicked two goals in a win against St Kilda. He managed to kick two goals once again in the next game, a 76-point loss to Carlton, although he also kicked two behinds as part of the wayward kicking by the entire Essendon team, which managed 9.19 for the night. Hardingham was delisted at the conclusion of the 2014 AFL season. He remained with Essendon as a VFL-listed player in its reserves team in 2016. Hardingham, along with 33 other Essendon players, was found guilty of using a banned performance-enhancing substance, thymosin beta-4, as part of Essendon's sports supplements program during the 2012 season. He and his teammates were initially found not guilty in March 2015 by the AFL Anti-Doping Tribunal, but a guilty verdict was returned in January 2016 after an appeal by the World Anti-Doping Agency. He was suspended for two years which, with backdating, ended in November 2016; as a result, he served approximately fourteen months of his suspension and missed the entire 2016 season.

== History == Following World War II, Sonic founder Troy N. Smith Sr., returned to his hometown of Seminole, Oklahoma, where he became employed as a milkman. He then decided to work delivering bread, because bread is lighter than milk. Soon afterwards, Smith purchased the Cottage Cafe, a little diner in Shawnee, Oklahoma. Before long, he sold it and opened a fast-food service, Troy's Pan Full of Chicken, on the edge of town. In 1953, Smith joined with a business partner to purchase a five-acre parcel of land that had a log house and a walk-up root beer stand named the Top Hat. The two continued operating the root beer stand and converted the log house into a steak restaurant. After realizing that the stand was averaging $700 a week in the sale of root beer, hamburgers, and hot dogs, Smith decided to focus on the more-profitable root beer stand. He bought out his business partner. Initially, Top Hat customers parked their automobiles anywhere on the gravel parking lot and walked up to place orders. In Louisiana, Smith saw a drive-in that used speakers for ordering. He suspected that he could increase his sales by controlling the parking and having the customers order from speakers at their cars, with carhops delivering the food to the cars. Smith borrowed several automobiles from a friend who owned a used-car lot to establish a layout for controlled parking. He also had some so-called "jukebox boys" wire an intercom system in the parking lot. Sales immediately tripled. Charles Woodrow Pappe, an entrepreneur, was impressed by the Shawnee drive-in.

The principal difference is a chemical cap on the N-terminal tyrosine that prevents rapid enzymatic cleavage. Native GHRH is degraded within minutes in plasma, whereas the modified peptide persists considerably longer. The amino acid backbone otherwise mirrors the natural hormone.
