Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from truncated or oxidized forms. Mass spectrometry confirms the expected molecular weight, and peptide mapping after enzymatic digestion verifies the amino acid sequence. Water content is measured because residual moisture affects stability, and tests for aggregates or particulates are standard for injectable peptides. Circular dichroism can indicate whether the molecule has adopted an unexpected secondary structure in solution.
Research supply is often accompanied by a certificate of analysis listing chromatographic purity, mass confirmation, and storage conditions. Laboratories compare that document with an independent test when material is intended for bench work, since certificates describe a batch rather than an individual vial. Published studies usually state the source and purity of the peptide because small differences in purity can shift measured activity. Full analytical validation is rarely reported, which leaves batch-to-batch comparability an open question.
Questions remain about how much of the observed fat reduction reflects direct GHRH-receptor signaling versus the downstream growth hormone and IGF-1 surge. It is also unclear whether the compound produces meaningful benefit in populations without lipodystrophy, since trials in cognitive impairment did not reach their stated goals. Long-term effects on glucose metabolism and on cardiovascular outcomes are not fully characterized. Published work generally describes effects on surrogate markers rather than on hard clinical endpoints, and independent replication of some findings is limited.
Tesamorelin acts on the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in intracellular cyclic AMP, which in turn opens ion channels and raises calcium concentrations, leading to release of stored growth hormone into the bloodstream. Because the peptide works through the same receptor as the body's own GHRH, the resulting secretion follows a pulsatile pattern rather than a continuous elevation. The N-terminal modification slows enzymatic breakdown, so the signal persists longer than it would with the unmodified hormone.

Pharmacodynamic studies show that tesamorelin reduces visceral adipose tissue more than subcutaneous adipose tissue in the studied population. This selectivity may relate to differences in blood flow and hormone sensitivity between fat depots. Effects on glucose metabolism and insulin sensitivity have been investigated, with some trials reporting modest changes and others showing stability. The precise relationship between growth hormone exposure, IGF-1 levels, and visceral fat loss remains an active area of analysis.
Tesamorelin binds to growth hormone-releasing hormone receptors on somatotroph cells in the anterior pituitary. Receptor activation increases intracellular cyclic AMP and promotes synthesis and secretion of growth hormone. Because the peptide mimics endogenous GHRH, it amplifies the normal pulsatile release of growth hormone rather than providing exogenous growth hormone directly. This upstream action distinguishes tesamorelin from recombinant growth hormone preparations and from growth hormone secretagogues that act at different receptors.
Storage claims vary across suppliers, and published stability data for specific formulations are limited. Extrapolating from related peptides is common but not a substitute for direct measurement. For research use, documentation such as a certificate of analysis is often requested to confirm identity and purity. What constitutes an acceptable purity threshold depends on the intended application. Open questions remain about how temperature excursions during shipping affect long-term peptide integrity. Independent verification by an end user is not routinely reported.
Lyophilized tesamorelin is generally stored refrigerated at 2 to 8 degrees Celsius, protected from light and moisture. Peptides in this class are often kept frozen at minus 20 degrees Celsius for longer periods. Reconstituted solutions are typically used within a defined window because hydrolysis and oxidation proceed faster in liquid form. Container material and headspace also influence how long a preparation retains its expected profile. Specific stability figures depend on concentration and buffer composition.
Common analytical approaches include reversed-phase high-performance liquid chromatography for purity assessment and mass spectrometry for identity confirmation. Peptide mapping after enzymatic digestion can verify the expected sequence. Immunoassays may be used to measure the compound or its downstream markers, but they can cross-react with related peptides and require careful validation. Impurity profiles typically include truncated sequences, oxidized methionine residues, and residual solvents from synthesis. Each method reports a different property, so no single assay establishes overall quality.

Robert William Barker (December 12, 1923 – August 26, 2023) was an American media personality, game show host, and animal rights advocate. He hosted CBS's The Price Is Right, the longest-running game show in North American television history, from 1972 to 2007. Barker also hosted Truth or Consequences from 1956 to 1975. Born in Darrington, Washington, in modest circumstances, Barker spent most of his youth on the Rosebud Indian Reservation and was a citizen of the Rosebud Sioux Tribe. Barker joined the United States Navy Reserve during World War II. He worked part-time in radio while attending college. In 1950, Barker moved to California to pursue a broadcasting career. He was given his own radio show, The Bob Barker Show, which ran for six years. Barker began his game show career in 1956, hosting Truth or Consequences. Barker began hosting The Price Is Right in 1972. He became an advocate for animal rights and of animal rights activism, supporting groups such as the United Activists for Animal Rights, People for the Ethical Treatment of Animals, and the Sea Shepherd Conservation Society. In 2007, Barker retired from hosting The Price Is Right after celebrating his 50-year career on television. Regarded as a pop culture icon, Barker continued to make occasional appearances for several years into his retirement until 2015.
=== Penicillin G === Penicillin G is destroyed by stomach acid, so it cannot be taken by mouth, but doses as high as 2.4 g can be given (much higher than penicillin V). It is given by intravenous or intramuscular injection. It can be formulated as an insoluble salt, and there are two such formulations in current use: procaine penicillin and benzathine benzylpenicillin. When a high concentration in the blood must be maintained, penicillin G must be administered at relatively frequent intervals, because it is eliminated quite rapidly from the bloodstream by the kidney. Penicillin G is licensed for use to treat septicaemia, empyema, pneumonia, pericarditis, endocarditis and meningitis caused by susceptible strains of staphylococci and streptococci. It is also licensed for the treatment of anthrax, actinomycosis, cervicofacial disease, thoracic and abdominal disease, clostridial infections, botulism, gas gangrene (with accompanying debridement and/or surgery as indicated), tetanus (as an adjunctive therapy to human tetanus immune globulin), diphtheria (as an adjunctive therapy to antitoxin and for the prevention of the carrier state), erysipelothrix endocarditis, fusospirochetosis (severe infections of the oropharynx, lower respiratory tract and genital area), Listeria infections, meningitis, endocarditis, Pasteurella infections including bacteraemia and meningitis, Haverhill fever; rat-bite fever and disseminated gonococcal infections, meningococcal meningitis and/or septicaemia caused by penicillin-susceptible organisms and syphilis.
The Gam-COVID-Vac vaccine was developed by a cellular microbiologists team of the government-backed Gamaleya Research Institute of Epidemiology and Microbiology. The group was led by MD and RAS associate member Denis Logunov, who also worked on vaccines for the Ebolavirus and the MERS-coronavirus. In May 2020, the Gamaleya Research Institute of Epidemiology and Microbiology announced that it had developed the vaccine without serious side effects. By August 2020, phases I and II of two clinical trials (involving 38 patients each) were completed. Only one of them used the formulation which later obtained marketing authorization under limited conditions. This vaccine was given the trade name "Sputnik V", after the world's first artificial satellite. During preclinical and clinical trials, 38 participants who received one or two doses of the Gam-COVID-Vac vaccine had produced antibodies against SARS-CoV-2's spike protein, including potent neutralizing antibodies that inactivate viral particles. On 11 August 2020, the Russian minister of Health Mikhail Murashko announced at a government briefing with the participation of President Vladimir Putin regulatory approval of the vaccine for widespread use. The state registration of the vaccine was carried out "conditionally" with post-marketing measures according to the decree of the Government of the Russian Federation.
On 14 January 2015, a landmark ruling was made by the Court of Appeal in the prosecution's appeal against the re-sentencing case of one former death row inmate, Kho Jabing, who was re-sentenced to life imprisonment and 24 strokes of the cane for the murder of Chinese national Cao Ruyin during a robbery under Section 300(c) of the Penal Code of Singapore. The landmark judgement in which the court, by a majority decision of 3–2, overturned Kho's life sentence and sentenced him to death a second time, had set the main guiding principles for all judges in Singapore to decide if the death penalty is appropriate for those murder cases committed with no intention to kill while exercising their discretion to impose either a life term or death for offenders responsible for such. The main guiding principles set were as such:
Off California, white sharks will eat cabezons, white seabasses, lingcod, halibut, leopard sharks, smooth-hounds, spiny dogfishes, school sharks, stingrays, bat rays, and skates. In the Mediterranean, they consume Atlantic bluefin tunas, bullet tunas, Atlantic bonitos, swordfishes, blue sharks, shortfin makos, and stingrays. An ocean sunfish was also recorded in a white shark's stomach. Off the northeastern US, juveniles commonly eat bottom-dwelling fish like hake, while off South Africa, they often prey on dusky sharks. The remains of an adult whale shark were found in a white shark, though whether this was active hunting or scavenging could not be determined. They are also recorded to consume cephalopods, as evidenced by beaks found in their stomachs. Off South Africa, white sharks under 2.5 m (8.2 ft) were found with remains of coastal and bottom-dwelling species like certain octopus species, as well as species of the genera Sepia and Loligo, while larger sharks seem to prefer more open-ocean species like those of the genera Ancistrocheirus, Octopoteuthis, Lycoteuthis, Ornithoteuthis, Chiroteuthis, and Argonauta. Near Guadalupe, white sharks have been documented with scars that appear to have been caused by neon flying squids, jumbo squids, and giant squids. Both fish and cephalopods may be important food sources at the White Shark Café. Other animals recorded as prey include sea turtles.

The pump sends the oil through the hole in the lower part of the gauge. Saw bar producers provide a large variety of bars matching different saws. Grease is pumped through a hole at the bar nose, typically each tank filling to keep the nose sprocket well lubricated. One or two bolts from the saw run through a guide slot. The clutch cover is put on top of the bar and it is secured through these bolts. The number of bolts is determined by the size of the saw. Different bar types are available:

== History == In the early 1960s, Dr. Arnold Kadish, a physician working at Loma Linda University, developed a closed-loop device for blood glucose control using a double-lumen autoanalyzer to measure blood glucose linked to an intravenous insulin infusion pump. Although it worked, the prototype was considered so bulky, worn as a backpack, and complicated, that it was never commercialized. In 1976, the first commercialized insulin pump was created and was named the Biostator. The pump was a 60 kg bedside device. It also had the capability of monitoring blood glucose levels, so this also doubles as the first continuous glucose monitor. Between 1978 and 1988, Robert Channon, working with Guy's Hospital and the Bristol Royal Infirmary, developed a series of miniature insulin infusion pumps. Today, insulin pumps are so small that they can fit in a pocket or a purse. In the late 1970s and early 1980s, Sandia National Laboratories and the University of New Mexico School of Medicine developed a remotely programmable implantable insulin infusion system that was tested in animals and implanted in human patients beginning in 1981. In 1984, an Infusaid implantable infusion device was used to treat a 22-year-old patient successfully. The insulin pump was first endorsed in the United Kingdom in 2003 by the National Institute for Health and Care Excellence (NICE).

Marxism–Leninism–Maoism became the name for the ideology of the Chinese Communist Party and of other Communist parties, which broke off from national Communist parties, after the Sino–Soviet split, especially when the split was finalised by 1963. The Italian Communist Party was mainly influenced by Antonio Gramsci, who gave a more democratic implication than Lenin's for why workers remained passive. A key difference between Maoism and other forms of Marxism–Leninism is that peasants should be the bulwark of the revolutionary energy, which is led by the working class. Three common Maoist values are revolutionary populism, pragmatism, and dialectics. According to Rachel Walker, "Marxism–Leninism" is an empty term that depends on the approach and basis of ruling Communist parties, and is dynamic and open to redefinition, being both fixed and not fixed in meaning. As a term, "Marxism–Leninism" is misleading because Marx and Lenin never sanctioned or supported the creation of an -ism after them, and is revelling because, being popularized after Lenin's death by Stalin, it contained three clear doctrinal and institutionalized principles that became a model for later Soviet-type regimes; its global influence, having at its height covered at least one-third of the world's population, has made Marxist–Leninist a convenient label for the Communist bloc as a dynamic ideological order.

Gonadotropin receptors are embedded in the surface of the target cell membranes and coupled to the G-protein system. Signals triggered by binding to the receptor are relayed within the cells by the cyclic AMP second messenger system. Gonadotropins are released under the control of gonadotropin-releasing hormone (GnRH) from the arcuate nucleus and preoptic area of the hypothalamus. The gonads — testes and ovaries — are the primary target organs for LH and FSH. The gonadotropins affect multiple cell types and elicit multiple responses from the target organs. As a simplified generalization, LH stimulates the Leydig cells of the testes and the theca cells of the ovaries to produce testosterone (and indirectly estradiol), whereas FSH stimulates the spermatogenic tissue of the testes and the granulosa cells of ovarian follicles, as well as stimulating production of estrogen by the ovaries. Although gonadotropins are secreted in a pulsatile manner (as a result of pulsatile GnRH release), unlike the case of GnRH and GnRH agonists, constant/non-pulsatile activation of the gonadotropin receptors by the gonadotropins does not produce functional inhibition. This can be seen during the first 7–10 weeks of pregnancy, where constantly high and progressively-increasing levels of hCG circulate and mediate production of estrogen and progesterone by the corpus luteum until the placenta takes over the production of these hormones.
== Research == New biologic therapies that target both existing cellular targets (including IL-12 and IL-23) and new cellular targets are being developed. Brazikumab and risankizumab are both IL-23 specific antagonists, opposed to ustekinumab which targets both IL-12 and IL-23, that have shown efficacy in phase 2 trials for Crohn's disease. Etrolizumab is an integrin receptor antagonist that targets beta 7 integrins. Etrolizumab has shown efficacy in phase 2 trials as well. The hope is that etrolizumab can show similar efficacy to natalizumab while avoiding the specific cellular target that is believed to have caused the instances of progressive multifocal leukoencephalopathy. Another area of research is focusing on the personalization of biological therapy. The idea is to use a specific patient's biochemical or genetic profile to predict how a patient will respond to a biological therapy. The information could help inform which class of biologics to use first. Personalized medicine is already being used in practice in the oncology field. A lot of research is being done to develop a biologic that can be delivered orally to address the many drawbacks associated with systemic administration. The general consensus in the field is that oral delivery of biologics directly to the diseased tissue could greatly reduce side effects, the development of anti-drug antibodies, and the cost of treatment.
== Advantages == There are many advantages to using a proton beam over an electron beam. There is less crystal charging from Bremsstrahlung radiation, although there is some from the emission of Auger electrons, and there is significantly less than if the primary beam was itself an electron beam. Because of the higher mass of protons relative to electrons, there is less lateral deflection of the beam; this is important for proton beam writing applications.

=== Theories of origin === The most commonly held of the three theories of tofu's origin maintains that tofu was discovered by Liu An (179–122 BC), a Han dynasty prince. While plausible, the paucity of reliable sources for this period makes this difficult to conclusively determine. In Chinese history, important inventions were frequently attributed to important leaders and figures of the time. It was used as a cheaper option to meat. In 1960, a stone mural unearthed from an Eastern Han dynasty tomb provided support for the theory of the Han origin of tofu; however some scholars maintain that tofu during the Han dynasty was rudimentary and lacked the firmness and taste for it to be considered as tofu. Another theory suggests that the production method for tofu was discovered accidentally when a slurry of boiled, ground soybeans was mixed with impure sea salt. Such sea salt would probably have contained calcium and magnesium salts, allowing the soy mixture to curdle and produce a tofu-like gel. The last group of theories maintains that the ancient Chinese learned the method for curdling soy milk by emulating the milk curdling techniques of the Mongolians. The primary evidence for this theory is the etymological similarity between the Chinese term rǔfǔ (乳腐), which literally means "milk curd", used during Sui dynasty (AD 581–618), for dishes with a consistency like yogurt or soft cheese, later influenced by Mongolian milk products and methods of production, and the term dòufu (豆腐, "bean curd") or tofu.

Lyophilized material is typically kept refrigerated and away from light in the sealed vial provided. Dissolved material is generally used within a limited period rather than stored long term.
Mass spectrometry gives the molecular weight, and mapping after digestion gives sequence coverage. Reversed-phase chromatography then supplies a purity profile.
Different groups report purity with different methods and thresholds, and full validation data are seldom published. Direct comparison of activity across batches therefore stays uncertain.
It acts upstream at the pituitary receptor and depends on functioning somatotroph cells to produce any effect. Growth hormone injections bypass that step and deliver the hormone directly. The pharmacokinetic profiles and the resulting feedback on the body's own secretion therefore differ.
