A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.

Regulatory approval in the United States came in 2010, when the Food and Drug Administration cleared the peptide for the reduction of excess abdominal fat in adults with HIV infection and associated lipodystrophy. The decision rested mainly on two randomized phase 3 trials that enrolled roughly eight hundred patients and ran for twenty-six weeks. Participants receiving active drug showed substantially greater declines in visceral adipose tissue than those receiving placebo, while total body weight changed comparatively little. A reformulated presentation was later approved, and the product has remained a niche therapy rather than a general weight-loss agent.
Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.
Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.
Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
Clinical investigation has focused on HIV-associated lipodystrophy, a condition in which antiretroviral therapy contributes to abnormal fat distribution. Excess visceral adipose tissue accumulates in the abdomen while peripheral fat may be lost. Tesamorelin was evaluated for reducing this visceral fat depot, with trials measuring changes in abdominal fat by imaging rather than by body weight alone. The rationale rests on the known lipolytic effects of growth hormone. Effects on visceral fat are documented, while long-term outcomes regarding cardiovascular risk remain less clearly established.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, composed of 44 amino acids. It was designed to retain the biological activity of the native hormone while resisting rapid enzymatic degradation. The compound is classified as a growth hormone secretagogue and belongs to the broader family of hypothalamic releasing factors. In research and clinical settings, it is studied for its ability to stimulate pituitary growth hormone release. Its structure includes a modification at the N-terminus that contributes to an extended half-life relative to native growth hormone-releasing hormone.
==== Productivity and adoption rates ==== While experimental studies show substantial productivity improvements for workers assisted by AI, such as software developers completing tasks 26% faster and mid-level professionals spending 40% less time on routine writing, these gains have not yet translated into economy-wide productivity growth in the UK. McKinsey & Company describes this as the "new productivity paradox", noting that over the past year, the largest contributions to UK productivity growth came from sectors with relatively low AI exposure, such as retail and facilities management. This paradox is partly explained by adoption patterns. A March 2026 survey by the British Chambers of Commerce (BCC) found that while AI adoption among SMEs had accelerated to 54% (up from 25% in 2024), most firms were using generic tools (like chatbots) rather than deeply integrated, bespoke systems. Furthermore, a 2026 global study by PwC showed that nearly 74% of AI's economic value is being captured by just 20% of organisations, being those that use AI to reinvent business models and pursue growth, rather than merely seeking cost reductions.
The Victor Babeș Clinical Hospital for Infectious Diseases and Pneumophthisiology (Romanian: Spitalul Clinic de Boli Infecțioase și Pneumoftiziologie „Victor Babeș”) was founded in 1925 in the Green Forest of Timișoara as the only infectious diseases hospital in western Romania. It bears the name of Romanian pathologist Victor Babeș since 1952.
== Selectivity == In the competition of bacterial cells and host cells with the antimicrobial peptides, antimicrobial peptides will preferentially interact with the bacterial cell to the mammalian cells, which enables them to kill microorganisms without being significantly toxic to mammalian cells. With regard to cancer cells, they themselves also secrete human antimicrobial peptides including defensin, and in some cases, they are reported to be more resistant than the surrounding normal cells. Therefore, we cannot conclude that selectivity is always high against cancer cells.
==== United Kingdom ==== Prescriptions of Oxycodone rose in Scotland by 430% between 2002 and 2008, prompting fears of usage problems that would mirror those of the United States. The first known death due to overdose in the UK occurred in 2002.
21 October – Stephen Flynn, the SNP's leader at Westminster, tables a motion urging the UK government to formally strip Prince Andrew of his royal titles following allegations made against him in the memoirs of Virginia Giuffre. A third, unnamed member of the grooming gang inquiry panel, resigns. Reform UK elects a new leader and deputy leader on Cornwall Council. 22 October – Angela Rayner gives her formal resignation speech to the House of Commons after leaving the frontbench. Jim Gamble becomes the second chair of the grooming gang inquiry to resign, while a fourth member resigns from the inquiry's panel as Starmer tells Prime Minister's Questions that the inquiry will look at racial and religious motives. King Charles and Queen Camilla arrive in Rome ahead of a historic state visit to Vatican City, where they will meet Pope Leo XIV. Reform UK's Linden Kemkaran issues a statement calling for councillors to back her in writing following advice to do so from her party's headquarters. 3 Labour members of Swindon Borough Council defect to the Green Party of England and Wales. 23 October – Lindsay Whittle wins the 2025 Caerphilly by-election for Plaid Cymru, taking the seat from Labour, who had held it since the Senedd was established in 1999. The UK government says it will not give MPs parliamentary time to debate whether Prince Andrew should be stripped of his titles. 24 October – 3 Scottish Green councillors defect to Jeremy Corbyn's Your Party, giving the party its first Scottish representatives.

The president is directly elected by popular vote for a renewable six-year term. Requirements for this position, which were enacted by the PAP government, are extremely stringent, such that only a handful of people qualify for the candidacy. These qualifications include that a candidate needs to be a person at least 45 years of age who is no longer a member of a political party, to either have held public office for at least 3 years in a number of specific public service leadership roles, or to have 3 years experience as chief executive of a fully profitable private sector company with at least S$500 million in shareholders' equity, be a resident in Singapore for at least 10 years, not have a criminal record, and more. Candidates must also "satisfy" the Presidential Elections Committee (PEC) that they are a person of integrity, good character and reputation. From 2017, the Constitution requires that presidential elections be "reserved" for a racial community if no one from that ethnic group has been elected to the presidency in the five most recent terms. Only members of that community may qualify as candidates in a reserved presidential election. In the 2017 presidential election, this combination of stringent requirements and a reserved election that required the candidate to be of the 13% Malay ethnic group led to the PEC approving a single candidate for the presidency; Halimah Yacob, considered part of the Malay community, won in an uncontested election. She also became Singapore's first female president.
== Indications (biomarkers) == The most common biomarker for exposure to AzM is the inhibition of AChE. Also other esterase enzymes as CaE and BChE are inhibited by AzM. In general AzM exposure can be better detected by AChE inhibition than CaE inhibition. In amphibians and also zebrafish, AChE is a more sensitive biomarker for low AzM exposure-levels. As already mentioned in paragraph 7 “detoxification”, AzM can be metabolized into nontoxic dimethylated alkylphosphates (AP), with the help of CYP450 and glutathione. These APs are: dimethylphosphate (DM), dimethylthiophosphate (DMTP) and dimethyldithiophosphate (DMDTP). These three metabolites may be excreted into the urine and can be used as reliable biomarkers of exposure to AzM. However these metabolites are not specific to AzM, because other organophosphate pesticides might also be metabolized into the three alkylphosphates. The amount of erythrocyte acetylcholinesterase (RBE-AChE) in the blood can also be used as a biomarker of effect for AzM. According to Zavon (1965) RBC-AChE is the best indicator of AChE activity at the nerve synapse, because this closely parallels the level of AChE in the CNS and PNS. A depression of RBC-AChE will correlate with effects due to a rapid depression of AChE enzymes found in other tissues, this is due to the fact that both enzymes can be inhibited by AzM.
==== Acquisitions and mergers ==== In June 2000, Pfizer acquired Warner-Lambert outright for $116 billion. To satisfy conditions imposed by antitrust regulators at the Federal Trade Commission, Pfizer sold off or transferred stakes in several minor products, including RID (a shampoo for treatment of head lice, sold to Bayer) and Warner-Lambert's antidepressant Celexa (which competes with Zoloft). The acquisition created what was, at the time, the second-largest pharmaceutical company worldwide. In 2003, Pfizer merged with Pharmacia, and in the process acquired Searle and SUGEN. Searle had developed Flagyl (metronidazole), a nitroimidazole antibiotic medication used particularly for anaerobic bacteria and protozoa. Searle also developed celecoxib (Celebrex) a COX-2 inhibitor and nonsteroidal anti-inflammatory drug (NSAID) used to treat the pain and inflammation in osteoarthritis, acute pain in adults, rheumatoid arthritis, ankylosing spondylitis, painful menstruation, and juvenile rheumatoid arthritis. SUGEN, a company focused on protein kinase inhibitors, had pioneered the use of ATP-mimetic small molecules to block signal transduction. The SUGEN facility was shut down in 2003 by Pfizer, with the loss of more than 300 jobs, and several programs were transferred to Pfizer. These included sunitinib (Sutent), a cancer medication which was approved for human use by the FDA in January 2006. A related compound, SU11654 (Toceranib), was also approved for cancer in dogs, and the ALK inhibitor Crizotinib also grew out of a SUGEN program.
=== Menstrual leave policy === In October 2025, the Karnataka cabinet approved a policy providing women employees with one paid menstrual-leave day per month, amounting to up to 12 days annually Lad described the measure as a progressive labour policy and said it was intended to support women's health and participation in the workforce. The policy was subsequently implemented through government orders. In December 2025, Karnataka extended the paid menstrual-leave benefit to women government employees, including contractual workers.

Genetic gingival enlargement presents in children with firm, fibrotic, generalized gingival overgrowth beginning around tooth eruption, often requiring surgical management. These cases are identified by tissue overgrowth-related pseudopocket formation and generally intact periodontal attachment.
Both copies of the affected gene must carry the same mutation (homozygous condition) for a person to be affected by an autosomal recessive disorder. An affected person usually has unaffected parents, each of whom carries one mutated gene and one normal gene (heterozygous condition) and is referred to as a genetic carrier; they may not have any symptoms. When both parents have the sickle cell trait, any given child has a 25% chance of sickle cell disease; a 25% chance of no sickle cell alleles, and a 50% chance of the heterozygous condition (see diagram). There are several different haplotypes of the sickle cell gene mutation, indicating that it may have arisen spontaneously in different geographic areas. The variants are known as Cameroon, Senegal, Benin, Bantu, and Saudi-Asian. These are clinically important because some are associated with higher HbF levels, e.g., Senegal and Saudi-Asian variants, and tend to have milder disease. The gene defect is a single nucleotide mutation of the β-globin gene, which results in the amino acid glutamic acid being substituted by valine at position 6 of the β-globin chain. Haemoglobin S with this mutation is referred to as HbS, as opposed to the normal adult HbA. Under conditions of normal oxygen concentration, this causes no apparent effects on the structure of haemoglobin or its ability to transport oxygen around the body. However, the deoxy form of HbS has an exposed hydrophobic patch, which causes HbS molecules to form long, inflexible chains.
In vertebrates, the nervous system consists of two main parts, the central nervous system (CNS) and the peripheral nervous system (PNS). The primary effect of leptins is in the hypothalamus, a part of the central nervous system. Leptin receptors are expressed not only in the hypothalamus but also in other brain regions, particularly in the hippocampus. Thus some leptin receptors in the brain are classified as central (hypothalamic) and some as peripheral (non-hypothalamic). As scientifically known so far, the general effects of leptin in the central nervous system are:
The white-minority National Party government were initially supportive, seeing SASO's creation as a victory for apartheid's ethos of racial separatism. Influenced by the Martinican philosopher Frantz Fanon, Biko and his compatriots developed Black Consciousness as SASO's official ideology. The movement campaigned for an end to apartheid and the transition of South Africa toward universal suffrage and a socialist economy. It organised Black Community Programmes (BCPs) and focused on the psychological empowerment of black people. Biko believed that black people needed to rid themselves of any sense of racial inferiority, an idea he expressed by popularizing the slogan "black is beautiful". In 1972, he was involved in founding the Black People's Convention (BPC) to promote Black Consciousness ideas among the wider population. The government came to see Biko as a subversive threat and placed him under a banning order in 1973, severely restricting his activities. He remained politically active, helping organise BCPs such as a healthcare centre and a crèche in the Ginsberg area. During his ban he received repeated anonymous threats, and was detained by state security services on several occasions. Following his arrest in August 1977, Biko was beaten to death by state security officers. Over 20,000 people attended his funeral. Biko's fame spread posthumously. He became the subject of numerous songs and works of art, while a 1978 biography by his friend Donald Woods formed the basis for the 1987 film Cry Freedom.
2,4,5-Trimethoxyphenethylamine (2,4,5-TMPEA; 2C-O), the 2C positional isomer of mescaline (3,4,5-trimethoxyphenethylamine), was first synthesized by Max Jansen and was reported to produce psychedelic effects similar to those of mescaline in 1931. However, subsequent studies in the 1960s and 1970s suggested that 2,4,5-TMPEA may actually be inactive as a psychedelic in animals and humans. 2C-D was the first of the 2C drugs after 2C-O to be discovered. It was synthesized and studied in animals by Beng T. Ho and colleagues at the Texas Research Institute of Mental Sciences and they published their findings in 1970. Alexander Shulgin synthesized 2C-B and 2C-D in 1974 and discovered their psychedelic effects in self-experiments conducted in 1974 and 1975. He published his findings in the scientific literature in 1975. However, Shulgin had previously tested sub-threshold doses of 2C-D in 1964 and 1965. 2C-T was first described by Shulgin and David E. Nichols in 1976. 2C-I was first described by Shulgin and colleagues in 1977 and initial psychoactivity was reported by Shulgin in 1978. Shulgin also first synthesized 2C-E in 1977. He reviewed several of these 2C drugs in a literature review in 1979. Subsequently, numerous other 2C drugs have been synthesized and characterized. Shulgin comprehensively reviewed and described the 2C drugs in his 1991 book PiHKAL (Phenethylamines I Have Known and Loved).
